Term Description
Detection of nucleic acid sequences specific to staphylococcal cassette chromosome mec (SCCmec), a genomic island found in methicillin-resistance Staphylococcus aureus (MRSA). This term was created for, but not limited in use to, Cepheid's Xpert MRSA/SA Nasal assay, a qualitative real-time PCR test that detects Staphylococcus aureus (SA) and methicillin-resistant Staphylococcus aureus (MRSA) DNA from nasal swabs in patients at risk for nasal colonization. Primers and probes are used to detect sequences of the staphylococcal protein A (spa), the gene for methicillin/oxacillin resistance (mecA), and the staphylococcal cassette chromosome (SCCmec) inserted into the Staphylococcus aureus chromosomal attB site.

SCCmec, or staphylococcal cassette chromosome mec, is a mobile genetic element of Staphylococcus bacterial species. This genetic sequence includes the mecA gene coding for resistance to the antibiotic methicillin and is the only known way for Staphylococcus strains to spread the gene in the wild by horizontal gene transfer.

Protein A is a 56 kDa MSCRAMM surface protein originally found in the cell wall of the bacterium Staphylococcus aureus.
As a pathogen Staphylococcus aureus utilizes Protein A, along with a host of other proteins and surface factors to aid its survival and, thus, virulence. Protein A helps inhibit phagocytic engulfment and acts as an immunological disguise. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Higher levels of Protein A in different strains of S. aureus have been associated with nasal carriage of this bacteria.
