72163-9 Leukocytes [Presence] in Specimen by Gram stain
Term Description
The qualitative assessment of leukocytes are recorded along with the presence of bacteria, yeast, and other cells from a Gram stain on any part of the body where infection is suspected. Example specimens include swab of abscess, cervical or vaginal swab, pus, throat or nose swab. Results are used for diagnosis, selection of antibiotics for treatment, and antimicrobial susceptibility testing.

Part Descriptions
LP14419-3 Leukocytes
Leukocytes or white blood cells (WBCs) are immune cells that fight infection, neoplasms and other inflammatory conditions, and mediate allergic responses. There are five types of WBCs normally present in the circulation that are all derived from a similar stem cell in the bone marrow. The five type of WBCs are divided into two groups based on the presence or absence of granules in the cytoplasm. The granulocytes include the neutrophils, basophils and eosinophils. The non-granulocytes include the lymphocytes and the monocytes. The neutrophils fight infection by ingesting and digesting bacteria. Eosinophils and basophils respond to allergic reactions and are capable of ingesting antigen-antibody complexes. Monocytes phagocytose bacteria and release interferon to stimulate the immune system. Lymphocytes are divided into T-cells and B-cells. T-cell immunity is cellular and involves the activation of phagocytes and B-cell immunity uses antibodies to fight infection. Both elevated and low leukocyte counts can be markers of infection and malignancy, and low leukocyte counts are associated with a variety of primary and secondary immunodeficiencies, depending on the WBC type(s) that are out of range. 14368-5 Trichomonas vaginalis [Presence] in Urethra by Wet preparation
Part Descriptions
LP14413-6 Trichomonas vaginalis
Trichomonas vaginalis (TV) is an anaerobic, flagellated parasitic protozoan that is among the most common non-viral sexually transmitted organisms whose only known host is humans. It is estimated that 170 million new cases occur worldwide and it is the most common sexually transmitted diseases in the United States. TV is known to increase HIV acquisition and transmission in both men and women by acting as a cofactor to increase susceptibility to HIV of an uninfected partner and infectivity of HIV of an infected partner. In women, infection with TV is a possible risk factor for developing cervical neoplasia, post-operative infections, adverse pregnancy outcomes, atypical pelvic inflammatory disease and infertility. In men, infection with TV can lead to non-gonococcal urethritis, infertility, chronic prostatitis and decreased sperm motility. Diagnosis is most often made using wet mount microscopy which has a sensitivity of only 52%. Other diagnostic methods include Pap smear, culture and PCR with sensitivities of 56%, 78% and 84%, respectively. There is an estimated 5 million new cases of TV annually in the United States and yet TV is still not a reportable disease. Treatment for TV is with metronidazole though emerging metronidazole-resistant strains have been reported. Asymptomatic infection can persist for months to years. LP6760-5 Wet preparation
A wet preparation or wet mount is used to identify the cause of vaginosis, a general term for infections of the vagina. A drop of vaginal discharge is placed on a slide with a drop of liquid -usually water--which is then examined microscopically as an unstained wet mount or as a Gram-stained slide. The refractive index of the water improves the image quality. Wet mounts generally cannot be stored since the water evaporates. Glycerol or immersion oil can be used instead of water. Bacterial vaginosis, candidiasis and trichomoniasis are common diseases which may be diagnosed through a wet preparation. [Lab Tests Online Conditions:vaginitis/start/2]

14371-9 Yeast [Presence] in Urethra by Wet preparation
Part Descriptions
LP29134-1 Yeast
Fungi are eukaryotic microorganisms which manifest as yeasts, molds or a combination. Yeasts start as individual cells that reproduce by budding. Molds, which are long filaments or hyphae, grow by apical extension. Both yeasts and molds are heterotrophic and digest their food externally by absorptive nutrition. Taxonomically, yeast are characterized by their size and shape, the presence or absence of capsules, conidiogenesis (the process of daughter cell formation), presence of sexual spores, the formation of pseudo-and true hyphae and physiologic data. Species are differentiated based on the ability to assimilate and ferment carbon sources, and to utilize nitrate as nitrogen. Some opportunistic fungal pathogens found in humans are dimorphic. They grow as mycelium in nature and reproduce as yeast in the body. One example is Candida which can quickly transform from yeast to the hyphal phase in humans; this process helps it invade host tissue. [NCBI Books:NBK8125]

10354-9 Bacteria identified in Urethra by Culture
Part Descriptions
LP14082-9 Bacteria
One of the three domains of life (the others being Eukarya and Archaea), also called Eubacteria. They are unicellular prokaryotic microorganisms which generally possess rigid cell walls and multiply by cell division. They exhibit three principal forms: round or coccal, rodlike or bacillary, and spiral or spirochetal. Bacteria can be classified by their response to oxygen (aerobic, anaerobic, or facultatively anaerobic) and by how they obtain their energy (chemotrophic, i.e. via a chemical reaction or phototrophic, via a light reaction). In addition, chemotrophs obtain their energy from chemicals, lithotrophic from inorganic compounds and organotrophic from organic compounds. Bacteria are also classified by the source carbon that they utilize: heterotrophic, from organic sources or autotrophic, from carbon dioxide. They can also be classified by whether or not they stain (based on the structure of their cell walls) with crystal violet dye: gram-negative or gram-positive.

LP6209-3 Culture
A culture is a laboratory procedure where a sample of tissue or fluid from a patient is placed in or on growth media (agar, broth) to induce the proliferation of potentially infectious agents (bacteria, fungi, viruses). The presence of growth is documented and the organisms are identified to determine whether or not a pathogen is present.
Once a pathogen is identified, specific testing techniques can be used to determine drug susceptibilities for assistance in treatment. [https://courses.lumenlearning.com/boundless-microbiology/chapter/culturing-bacteria/]

697-3 Neisseria gonorrhoeae [Presence] in Urethra by Organism specific culture
Part Descriptions
LP14316-1 Neisseria gonorrhoeae
Neisseria gonorrhoeae are gram-negative cocci usually seen in pairs with the adjacent sides flattened. Pili, filament-like appendages, extend from the cell surface and have a role in adherence. Gonorrhea is usually acquired by sexual contact. Men and women aged 15-29 have the highest incidence of gonorrhea. Number of sexual partners, sexual practices and preference, condom use and population mobility contribute to disease incidence. The most frequent sites are the cervix, urethra, rectum, pharynx and conjunctiva. The most common symptom in men is discharge that may be scanty and clear or cloudy to copious and purulent, and often dysuria. Asymptomatic men are an important reservoir for transmission. Endocervical infection is the most common form of infection in women. Ocular infections occur most commonly in newborns who are exposed in the birth canal and can lead to corneal scarring or perforation. Gonococcal bacteremia may lead to disseminated infection including pelvic inflammatory disease in as many as 15 percent of women, leading to an increased probability of infertility and ectopic pregnancy. While antibiotics have successfully treated gonorrhea for decades, N. gonorrhoeae has developed antimicrobial resistance to every drug used for treatment. The Centers for Disease Control and Prevention recommends combination therapy using two antimicrobials with different mechanisms of action (e.g., a cephalosporin plus azithromycin) to improve treatment efficacy and potentially slow the spread of resistance to cephalosporins. 21191-2 Chlamydia trachomatis DNA [Presence] in Urethra by NAA with probe detection
Part Descriptions
LP14298-1 Chlamydia trachomatis
Chlamydia trachomatis is one of three bacterial species in the genus Chlamydia, family Chlamydiaceae, class Chlamydiae. C. trachomatis has only been found living inside human cells . It causes urethritis , epididymitis, and proctitis in men and cervicitis, salpingitis, urethritis and proctitis in women. C. trachomatis has also been detected in some patients with temporomandibular joint (TMJ) disease. It may be treated with any of several antibiotics such as azithromycin, erythromycin or doxycyclinetetracycline. C. trachomatis was the first chlamydial agent discovered in humans. It comprises two human biovars: trachoma and lymphogranuloma venereum (LGV). Many, but not all, C. trachomatis strains have an extrachromosomal plasmid.
