Dihydrofolate reductase (DHFR) is an enzyme that changes dihydrofolate to tetrahydrofolate, required for the synthesis of purines, thymidylic acid, and certain amino acids. Due to its critical function, DHFR is found in all organisms. In humans it is encoded by the DHFR gene, and can be the target of drugs such as methotrexate used to treat certain neoplasms and inflammatory diseases. In bacteria, the bacterial DHFR enzyme is targeted by synthetic antibiotics such as trimethoprim. The chromosomal version of the bacterial gene (commonly folA) is sometimes overridden by trimethoprim-resistant versions present on mobile elements. Mutations of DHFR resulted in widespread resistance to trimethoprim. The dfrA class of genes are found to be the largest family of mobile trimethoprim antibiotic resistance genes.
