Hippuric acid (Gr. hippos, horse, ouron, urine), benzoyl glycocoll or benzoyl amidoacetic acid, C9 H9 NO3 or C6 H5 CONHCH2 CO2 H, is an organic acid found in the urine of horses and other herbivores. Its molecular weight is 179.175 and melting point range is 188-191ºC. High concentrations of hippuric acid can also indicate a toluene intoxication. It is excreted when many aromatic compounds, such as benzoic acid and toluene, are taken internally. Justus von Liebig in 1829 showed that it differed from benzoic acid, and in 1839 determined its constitution, while in 1873 Victor Dessaignes (Ann. 87, p. 325) synthesized it by the action of benzoyl chloride on zinc glycocollide. It is also formed by heating benzoic anhydride with glycine (Theodor Curtius, Ber., 1884, 17, p. 1662), and by heating benzamide with monochloroacetic acid. It crystallizes in rhombic prisms which are readily soluble in hot water, melt at 187°C. and decompose at about 240°C. Hippuric acid is biological renal marker used to monitor exposure to toluene or benzyl alchonol. Hippuric acid and methyl hippuric acid are the principal metabolities of toluene and xylene, respectively. Occupational exposure to these chemicals can be monitored by measuring urinary metabolite excretion.Typical excretion of hippuric acid in non-exposed patients is 1.0 g/g creatinine. Methyl hippuric acid is not found in non-exposed patients.
