Description
Thymidine synthase is the enzyme that catalyzes the methylation of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP) using the methyl donor 5,10-methylenetetrahydrofolate (CH2THF) as a cofactor. dTMP is critical for DNA repair and replication. Thymidine synthase has become the target for drugs used in the treatment of some cancers including the drug 5-fluorouracil (5-FU). The active metabolite of 5-FU combines with the nucleotide binding site of TS which prevents dUMP from being able to bind with TS. The decreased amount of dUMP leads to decreased cell replication.
