Description
Cells expressing CD45 (CD45+) in the absence of CD3 (CD3-) are used as an initial gating population for identifying natural killer (NK) cells. In addition to CD3-, a majority of NK cells are further gated on the expression of CD56 (neural cell adhesion molecule) and CD16 (low-affinity IgG Fc receptor-FcgammaRIII) cell surface markers. However, NK cells are not a homogeneous population since not all NK cells express the CD56 and CD16 markers uniformly. PMID: 17920947 NK cells that express CD56+ and are CD16- are called regulatory or cytokine secreting NK cells, whereas NK cells that express CD16+ and are CD56+/- (dim or negative) are referred to as cytotoxic NK cells. PMID: 19278419 NK cell deficiencies may be present in various primary (monogenic) immunodeficiencies (e.g.autoimmune lymphoproliferative syndrome (ALPS), familial hemophagocytic lymphohistiocytosis (FHL), Hermansky-Pudlak syndrome, Papillon-Lefevre syndrome, severe combined immunodeficiencies) or in isolated NK cell deficiency (e.g. absolute NK cell deficiency (ANKD), the classic NK cell deficiency (CNKD), and the functional NK cell deficiency (FNKD)).
