Ojo-Amaize, EA Lymphocyte Mitogen Stimulation. Lymphocyte mitogen stimulation assays are used for in vitro assessment of cellular immunity in patients with immunodeficiency, autoimmunity, infectious diseases, cancer and chemical-induced hypersensitivity reactions. Lymphocytes from healthy human subjects have receptors for mitogens such as the plant lectin concanavalin A (UCon A), pokeweed mitogen (PWM), the protein A component of Staphylococcus aureus Cowan strain I (SpA) and chemicals. Lymphocytes respond to these mitogens (substances that stimulate large numbers of lymphocytes without prior sensitization and in contrast to antigens do not require a sensitized host). Both B cells and T cells can be stimulated with mitogens. Inability of lymphocytes to respond to mitogens is a sign of impaired cell-mediated or humoral immunity.
There is a good correlation between CD4+ T-cell subset numbers and mitogenic response to PWM at different disease states in patients with HIV infection. [3H]-thymidine uptake is the gold standard for measuring lymphocyte mitogen stimulation.
However, a newer and more practical method utilizing supravital stains shows a good correlation with [3H]-thymidine uptake. Flow cytometry was also recently applied to analyze proliferation of cultured cells in response to stimulation with mitogen.
