Solute carrier organic anion transporter family member 1B1 is a protein that in humans is encoded by the SLCO1B1 gene [HGNC Gene ID: 2645]. Variations in the SLCO1B1 gene have been significantly associated with a marked increase in systemic exposure to simvastatin and risk of muscle toxicity. PMID: 24918167 The overall effect size and significance of this association are striking for simvastatin compared to other statins. Patients with variations in SLCO1B1 (a transporter of simvastatin) may require a lower simvastatin dose or an alternative statin as compared to patients who do not have this variant.
