Thymidine synthase is the enzyme that catalyzes the methylation of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP) using the methyl donor 5,10-methylenetetrahydrofolate (CH2THF) as a cofactor. dTMP is critical for DNA repair and replication. Thymidine synthase has become the target for drugs used in the treatment of some cancers including the drug 5-fluorouracil (5-FU). The active metabolite of 5-FU combines with the nucleotide binding site of TS which prevents dUMP from being able to bind with TS. The decreased amount of dUMP leads to decreased cell replication. Studies have shown that the gene amplification of TS in the tumor cells correlate to resistance to 5-FU. http://www.nature.com/nrc/journal/v3/n5/fig_tab/nrc1074_F2.html
